Leisure activity did not protect dopamine ageing

Tracking 181 healthy older adults over five years, dopamine D2-like receptor availability declined in the caudate and putamen, but education and self-reported physical, cognitive and social leisure activities showed negligible associations with those levels or changes. Neither factor moderated the link between receptor loss and cognitive decline.

Why it matters

The brain's dopamine system loses integrity with age, and that loss has been linked to declines in working memory, episodic memory and processing speed. Separately, education and an engaged lifestyle full of physical, cognitive and social leisure activity are widely promoted as protective for the ageing brain, often under the banner of cognitive reserve. What has been unclear is whether those protective factors act on dopamine integrity specifically — whether more educated or more active people preserve receptor availability better, or cope better with the same amount of loss. This study set out to test both possibilities directly.

What they did

The researchers used longitudinal data from the COBRA prospective cohort study, which included 181 older adults at baseline, and analysed it with Bayesian structural equation modelling. Dopamine D2-like receptor availability in the caudate and putamen was measured with [11C]raclopride positron emission tomography. Cognition was assessed across working memory, episodic memory and perceptual speed. Education and self-reported physical, cognitive and social leisure activities were also recorded. Three questions were posed: whether education or leisure related to baseline receptor availability or five-year change, whether changes in leisure activity tracked changes in receptor availability, and whether either factor moderated receptor-cognition change-change correlations.

What they found

Receptor availability declined over five years in both the caudate and the putamen, confirming the expected ageing pattern. The correlations between change in receptor availability and change in cognition were, however, weak overall. For both baseline levels and five-year changes in receptor availability, associations with education and with leisure activities were uniformly negligible or small and not strongly supported by the modelling. Neither education nor leisure activities moderated the change-change correlations between receptor availability and cognition. In short, the study found decline but no evidence that either putative protective factor influenced it or buffered its cognitive consequences.

Where it fits

This complicates a popular narrative in brain ageing. Education and leisure engagement remain associated with better cognitive outcomes in many studies, but this analysis suggests dopamine receptor integrity is probably not the route through which they act — at least not detectably over five years in this cohort. The weak receptor-cognition change correlations also temper how central dopamine loss should be considered in normal cognitive ageing. Because leisure activity was self-reported and the design is observational, real differences in activity could be underestimated, and the study cannot rule out effects on other brain systems or over longer periods.

What it means for you

This is a reason to be sceptical of the specific claim that staying busy and mentally engaged preserves your dopamine system. It is not evidence that education, exercise or social engagement are useless for the ageing brain — those benefits are documented elsewhere and this study simply did not find dopamine receptors to be the mechanism. It is also a reminder that self-reported leisure activity is a blunt measure. The practical upshot is interpretive: brain-ageing protection is likely to work through several routes, and this one particular route did not show up.
